The species are present in all the countries of South America except Chile. Currently, basics information’s about the accurate taxonomic data and current distribution of the species that compose the group as well current phylogenetic relationship still lacking, especially in regions such as Marajó Island and its Western portion. Freshwater stingray’s also presents a number of taxonomic complications where morphology shape of the disc as well the color patter in many cases are not informative to species level. Although Brazil has a significant number of taxonomists, there are still groups of poorly known species, which may have species that not have been described and which need identification, which requires time, resources and trained people for such activity. Therefore, the expansion of integrated studies with molecular data that can facilitate species identification process and solve taxonomic problems has been increasing over time. DNA barcoding is a fast, efficient method globally used to define and identify new species. The benefit of DNA barcoding technique not only includes taxonomic studies, but also phylogenetic and population genetics. The results of the present study we identify at least 3 species Potamotrygonidae family in the region of Portel city, Marajó Island: P. motoro, P. orbignyi e P. aireba. However, when comparing the sequences of the present study with those available in GenBank, we found that P. motoro is not a monophyletic species, which was recently redescribed and limited only the basin of Paraná-Paraguay River. Thus this species from Portel city is probably a new species and not yet described.
