The economy of the Amazon region, especially of the riverside communities, strongly depends on the resources from the rivers, mainly of the fisheries. However, giving the river area of the region, the control of the fishing in this region ends up being scarce. DNA barcoding is an ally in this task because it is a technique that uses short DNA sequences as molecular diagnostics for identification at the species level and this information are obtained in a very fast and cheap way. This study intends to evaluate the possible substitution of commercially fresh fish popular named of “filhote” (Brachyplatystoma filamentosum) with the use of DNA barcoding, and for which species are being substituted, pointing out their possible causes. 12 samples of B. filamentosum were collected from Breves fish market, Marajó Island and 12 samples from the city of Mocajuba from Tocantins River, totaling 24 samples. Total genomic DNA was extracted with the Wizard Genomics DNA extraction kit (Promega Inc.) and were migrated in 1% low melt agarose gel underwater electrophoresis to quantify their quality. From the extraction, amplifications were performed through the Polymerase Chain Reaction (PCR) of the Cytochrome Oxidase subunit I - COI gene.
