The specimens have a remarkable diversity in shape, size and lifestyle. This order houses fish popularly known as "pufferfish" belonging to the families, Diodontidae and Tetraodontidae. Pufferfish have few external characteristics useful for taxonomy and these are easily distorted when specimens are fixed in formaldehyde and preserved in ethanol, making the process of species recognition and classification difficult, therefore, complementary to systematic tools, such as the use of DNA, have been assisting researchers in investigating the presence of new molecular strains. DNA Barcoding, is a method for the rapid identification of any species, based on the extraction of a DNA sequence from a small sample of any organism utilizing the gene COI like an universal marker. The objective of the present study is, apply the barcodes approach to investigate which lineage of the Colomesus genus, occurs in the region of Breves, Marajó’s Island. Muscle tissue samples were taken from eight specimens of Colomesus asellus collected in Breves and compared to sequences from other locations. The results revealed the formation of three distinct clades with significant support value. The data obtained show the possible existence of a new lineage within the genus.
