A method for the identification of species based on the Polymerase Chain Reaction (PCR) of the 5S rDNA gene was used in the present study for the differentiation of South American flatfishes belonging to two families, the Achiridae and Paralichthyidae. The results of the analysis indicate the presence of species-specific banding patterns, and it was possible to discriminate the species based on a simple migration in agarose gel. This procedure was also used to verify intraspecific variation, which was inexistent in all species, with all the individuals of each taxon presenting the exact same banding pattern. The results of this study provide a rapid and precise method for the discrimination of the flatfish species of two families found in South America, from which few morphological and genetic studies are available.
